Abstract
The expanded PAM SpCas9 variant (xCas9) can recognize a broad range of PAM sequences including NG, GAA, and GAT.
- xCas9 offers the broadest PAM compatibility reported among Cas9 proteins active in mammalian cells.
- Applications of xCas9 include targeted transcriptional activation, gene disruption, and base editing.
- xCas9 exhibits much greater DNA specificity than the commonly used SpCas9.
- xCas9 demonstrates substantially lower off-target activity at all NGG target sites tested.
- Minimal off-target activity is observed when targeting genomic sites with non-NGG PAMs.
- The findings suggest that increased PAM compatibility does not compromise DNA specificity.
