Development and description of tiny biodegradable particles carrying exenatide using a coacervation method

Abstract

A PLGA concentration of 6% w/w resulted in elevated yields of > 50% for microspheres encapsulating exenatide acetate.

  • Formulation variables such as DCM: Si oil volume ratios and hold time were evaluated to optimize microsphere production.
  • The stability of microspheres was confirmed with hold times between 10 seconds to 2 minutes before immersion in a heptane bath.
  • In-vitro testing showed a low burst release rate of 2-6% over 24 hours.
  • Continuous release profiles of the encapsulated exenatide were observed for over 56 days, consistent with the expected performance of the commercial product.
  • These findings provide a framework for developing formulations that can encapsulate both generic and new peptides using coacervation.
