Safety and effectiveness of GLP-1 and GLP-1/GIP receptor drugs versus other treatments for weight loss and type 2 diabetes, adjusted for exposure, based on FDA reports of over 34,000 safety and 36,000 effectiveness cases

Updated

Abstract

Weight loss was greatest with GLP-1/GIP dual agonists.

  • Mortality risk rates for GLP-1/GIP receptor agonists were lower (RR: 0.1) compared to non-GLP agents (RR: 0.8) and other GLP-1 RAs.
  • Morbidity rates varied, with non-GLP agents showing an RR of 1.1, while T2D GLP-1 RAs had an RR of 0.9.
  • Postmarketing reports indicated a 5.4% occurrence of neoplasms with WM GLP-1 RAs, compared to 0.9% for non-GLP agents.
  • 71% of pivotal trials for these medications have been indexed in PubMed, suggesting a significant level of scrutiny.
  • Safety signals were noted to be limited due to small sample sizes in the analyzed studies.
