How Fam3a controls hormone-processing enzymes to regulate GLP-1 production in insulin cells through Nr4a2 and Foxa2

Abstract

Pancreatic active GLP-1 levels increased in Fam3a knockout mice compared to wild-type mice.

  • A significant negative correlation was observed between FAM3A and PCSK1 gene expression in human α-cells.
  • Knockdown of Fam3a in αTC1.9 cells led to increased expression of PC1/3 and higher GLP-1 production.
  • Nr4a2 was identified as a key downstream mediator of Fam3a, with its expression being regulated by Fam3a.
  • Foxa2, in complex with Nr4a2, negatively regulated PC1/3 expression and GLP-1 production.
  • Fam3a knockout mice exhibited improved glucose tolerance and increased plasma insulin levels in response to glucose.
