Frontiers in genome editing

Genome-edited sheep created by delivering prime editing proteins into fertilized eggs using electric pulses

Updated

Abstract

Editing efficiencies of up to 22.78% were achieved using prime editing in sheep fibroblasts.

  • Direct electroporation of a prime editing formulation into sheep zygotes resulted in gene editing efficiencies of 19.8% for the TBXT gene and 20.1% for the FecB gene.
  • No significant differences were observed in cleavage or blastocyst rates between electroporated and untreated zygotes.
  • After transferring 54 electroporated blastocysts, 14 live lambs were born, with intended genetic edits detected in five of them (35.71%).
  • A lamb edited at the TBXT locus exhibited a distinct short-tail phenotype, achieving an allele frequency of up to 50.97%.
  • No credible off-target editing was detected, indicating the specificity of this delivery method.
