Genome-wide activation map of surface protein levels in human helper T cells

Updated

Abstract

16% of activated genes significantly alter the expression of at least one surface protein in CD4 T cells.

  • A novel platform, SCITO-Perturb-seq, was developed to map the regulation of 201 surface proteins in human CD4 T cells.
  • Analysis involved 3.6 million human CD4 T cells to assess the effects of CRISPR activation on surface protein expression.
  • Five modules corresponding to known CD4 T cell states were identified through the analysis of perturbation effects.
  • These modules reveal coordinated regulation of surface proteins that are not co-expressed in unperturbed cells.
