[6]-Gingerol from ginger boosts insulin release and sugar uptake to improve high blood sugar in type 2 diabetic mice

Abstract

Treatment with [6]-Gingerol for 28 days significantly increased glucose-stimulated insulin secretion in Leprdiabetic mice.

  • [6]-Gingerol improved glucose tolerance and elevated plasma GLP-1 levels in treated mice.
  • The insulin secretion effect of [6]-Gingerol was regulated by pharmacological modulation of GLP-1 levels.
  • [6]-Gingerol treatment activated signaling pathways involving cAMP, PKA, and CREB in pancreatic islets, which are important for insulin secretion.
  • There was an upregulation of Rab27a GTPase and its effector Slp4-a in isolated islets, linked to insulin exocytosis.
  • Treatment enhanced glycogen synthase 1 activity and increased GLUT4 transporter levels in skeletal muscle membranes, facilitating glucose disposal.

ETHICS APPROVAL: The designed experimental protocol was approved by the Ethics Committee on Animal Research, North South University. It was designed following the "Revised Guide for the care and use of laboratory animals by American Physiological Society” [48]. All animals were treated humanely throughout the course of the experiments and maximum care was taken to minimize the pain of the experimental animals. All surgical procedures were carried under sodium pentobarbitone anesthesia. Maintenance of the anesthesia was continuously monitored at 15mins interval by “toe pinch”.
