Abstract

Pancreatic uptake of (68)Ga-labeled exendin-4 was decreased by up to 90% in healthy rats when coadministered with DO3A-exendin-4.

  • GLP-1R-specific tissue uptake was significantly reduced in diabetic rats compared to healthy controls, with a decrease of more than 80%.
  • GLP-1R occupancy in the pancreas of cynomolgus monkeys varied from 49% to 97% after administration of DO3A-exendin-4 at doses ranging from 0.15 to 20 μg/kg.
  • The findings indicate that pancreatic uptake of (68)Ga-DO3A-exendin-4 may be linked to specific receptor binding.
  • Selective destruction of β-cells is associated with decreased GLP-1R uptake in the pancreas.
