Changes in gut bacteria and body-wide inflammation linked to oral semaglutide delivered with SNAC

Abstract

SNAC treatment resulted in a 62% depletion of primary fermenters in Muribaculaceae and a 77% depletion in Bacteroidaceae compared to the control group.

  • Oral semaglutide (SEM) with SNAC achieved 0.4-1% bioavailability through gastric absorption.
  • Microbial α-diversity remained stable, while SNAC significantly altered β-diversity.
  • Predicted saccharolytic enzyme abundance and fecal butyrate concentrations decreased by 77% with SNAC and 75% with combined SEM-SNAC treatment.
  • Plasma analysis revealed a 70% increase in tumor necrosis factor-α (TNF-α) and an 85% decrease in brain-derived neurotrophic factor (BDNF) in SNAC-treated rats.
  • SNAC treatment was associated with increased liver weight and reduced caecum mass.
