Genes that help Epstein-Barr virus infect human B cells found using a genome-wide CRISPR screen

Updated

Abstract

A genome-wide CRISPR-Cas9 screen identified new factors essential for EBV B cell infection.

  • EBV infects 90% of the global population and is linked to several cancers and autoimmune diseases.
  • New host factors and signaling pathways critical for EBV B cell infection were discovered, in addition to known receptors.
  • Knockout of CD19, CD81, or KPNA1 significantly hindered the establishment of EBV infection.
  • CD19 was found to co-localize with EBV during early infection stages, indicating its role in supporting EBV uptake.
  • Depletion of KPNA1 affected EBV infection establishment but did not impact the production of infectious EBV from reactivated B cells.
