Development and Testing of Microneedles Made from Hyaluronic Acid and Aminoclay for Painless Delivery of Semaglutide in Obesity Treatment

Abstract

Sema-AC/HA10 microneedles showed a mechanical strength of 0.37 ± 0.020 N/needle.

  • The microneedles retained stable physicochemical and morphological properties at 25°C during storage.
  • Encapsulated semaglutide maintained its structural integrity within the microneedles.
  • Sema-AC/HA10 dissolved quickly upon skin insertion, which could enhance drug absorption through the skin.
  • This formulation significantly improved systemic drug exposure compared to free semaglutide.
  • In vivo testing indicated that Sema-AC/HA10 reduced blood glucose, HbA1c, total cholesterol, triglycerides, food intake, water consumption, and body weight in type 2 diabetic rats.
