Topical administration of a pH-gated hydrogel patch achieved >20% NLRP3 editing efficiency in wound tissue.

  • The hydrogel patch delivers CRISPR-Cas9 ribonucleoprotein complexes in response to infection.
  • NLRP3 editing efficiency was 57% in J774A.1 cells and 69% in L929 cells under laboratory conditions.
  • The patch allows for localized presentation of gene editing tools while minimizing off-target effects.
  • In a mouse model of Staphylococcus aureus-infected wounds, the patch reduced inflammatory cell infiltration.
  • The approach may enable more effective gene therapy for chronic inflammatory wounds.
