Abstract

HIF-1alpha is expressed in human normal and osteoarthritic chondrocytes under normoxic conditions.

  • Both normal and osteoarthritic chondrocytes show detectable levels of HIF-1alpha under normoxic conditions.
  • Hypoxic conditions (1% O2) for up to 16 hours can stabilize or increase HIF-1alpha expression in chondrocytes.
  • Treatment with TNF-alpha leads to increased HIF-1alpha protein levels in chondrocytes under normoxic conditions.
  • Inhibition of NFkappaB or p38 MAP kinase partially blocks the increase in HIF-1alpha expression induced by TNF-alpha.
  • The expression of HIF-2alpha mRNA is also detected in human chondrocytes for the first time.
