Low oxygen factor may increase prostaglandin production in joint cartilage cells affected by osteoarthritis

Abstract

Enhanced expression of the mPGES-1 gene was observed in osteoarthritic cartilage compared to healthy cartilage.

  • An increased number of osteoarthritic chondrocytes exhibited staining for mPGES-1.
  • Exposure to low oxygen levels (1% O2) resulted in significant up-regulation of mPGES-1 in primary chondrocytes.
  • Hypoxia was associated with increased synthesis of mPGES-1 and nuclear accumulation of the transcription factor HIF-1alpha.
  • Stimulation with dimethyloxaloylglycine promoted the expression of mPGES-1 by stabilizing HIF-1alpha protein levels.
  • Treatment with 2-methoxyestradiol reduced mPGES-1 synthesis, correlating with decreased HIF-1alpha activity.
