Abstract

Among 68,351 case reports associated with incretin-based therapies, 1,327 (1.941%) involved hepatobiliary adverse events.

  • DPP-4 inhibitors showed statistically significant associations with hepatobiliary events such as cholelithiasis and chronic cholecystitis.
  • GLP-1 receptor agonists had weaker associations, primarily linked to gallbladder and biliary diseases.
  • Subgroup analyses indicated stronger correlations for liraglutide and semaglutide among GLP-1 agonists.
  • Among DPP-4 inhibitors, sitagliptin, linagliptin, and vildagliptin were associated with higher rates of hepatobiliary events.
  • The pooled reporting odds ratio of 2.85 suggests a positive correlation between incretin therapies and hepatobiliary adverse events.
