Blocking NAMPT worsens high-fat diet liver fat buildup in mice by affecting energy and fat-making signals

Abstract

Inhibition of NAMPT significantly promoted liver steatosis in mice fed a high fat diet.

  • Inhibition of NAMPT led to increased expression of lipogenic genes such as SREBP1 and fatty acid synthase (FASN).
  • Nicotinamide mononucleotide (NMN) and NAD were found to counteract the effects of NAMPT inhibition in liver cells.
  • Overexpression of NAMPT reduced hepatic lipid accumulation in liver cells.
  • Inhibition of NAMPT decreased protein levels of Sirt1 and phospho-AMPKα in the livers of high fat diet-fed mice.
  • Activation of Sirt1 with Resveratrol reduced lipogenic gene expression, whereas the specific Sirt1 inhibitor EX-527 had the opposite effect.
