  • Metformin is the most established glucose-lowering medication for diabetes prevention in selected high-risk individuals.
  • SGLT2 inhibitors offer cardiovascular and renal protection but lack sufficient evidence for routine use in diabetes prevention.
  • GLP-1 receptor agonists and tirzepatide have shown significant weight loss and cardiometabolic benefits that may help reduce T2DM progression.
  • Next-generation incretin-based therapies, such as triple receptor agonists, are still under investigation for their potential in T2DM prevention.
