Abstract

The multivalent aptamer system achieved a capture efficiency of 84.3%, significantly outperforming monovalent aptamers.

  • Subpopulation-specific miRNA profiling was performed using an integrated single-tube platform.
  • The new method combined EVs capture and miRNA analysis to enhance reproducibility.
  • In situ lysis of captured EVs increased local concentration of released miRNAs, improving signal detection.
  • A limit of detection as low as 1.42 × 10 particles/μL was achieved.
  • Distinct subpopulations of EVs displayed cell-line-specific expression patterns and heterogeneity.
  • The MUC1 subpopulation showed better clinical diagnostic performance than CD63 for miR-21 and miR-155.
