Using a temperature-sensitive nanoparticle to deliver Semaglutide through the nose: development, testing, and biological evaluation

Abstract

Nanoparticles containing semaglutide exhibited an average size of 26.14 nm and improved nasal absorption compared to the free drug.

  • The nanoparticle formulation achieved high encapsulation of semaglutide at 89% and showed enhanced stability against enzymatic degradation.
  • In vitro tests indicated increased permeation across nasal epithelial cells for the nanoparticle formulation compared to semaglutide alone.
  • Nasal cast studies demonstrated greater deposition of nanoparticles in the nasal turbinates versus the free drug.
  • In vivo studies revealed prolonged retention of the nanoparticle formulation in the sinus region and improved glucose tolerance.
  • No detectable brain fluorescence was observed, suggesting limited central nervous system exposure.
