Measuring Tirzepatide levels in rat blood to support drug behavior studies

Abstract

A liquid chromatography-tandem mass spectrometry method was developed for the quantification of tirzepatide in rat plasma with a linearity range of 1-1000 ng/mL.

  • Tirzepatide is a dual agonist targeting GIP and GLP-1 receptors.
  • The method demonstrated good accuracy and precision, meeting regulatory criteria across all quality control levels.
  • The terminal half-lives of tirzepatide were 10.04 hours after intravenous administration and 9.803 hours after subcutaneous administration.
  • Estimated bioavailability following subcutaneous dosing was approximately 62.38%.
  • The developed method may be applicable for quantitative analysis of other similar peptide therapeutics.
