Leucine supplementation increased perirenal adipose tissue by 27% in rats.

  • 5-week leucine supplementation reduced insulin signaling markers in rats, including a 36% decrease in tyrosine phosphorylation of insulin receptor β and IRS1.
  • Despite decreased insulin signaling, muscle glucose uptake in response to insulin did not show resistance in leucine-supplemented rats.
  • Overall glucose tolerance was impaired in rats receiving leucine supplementation.
  • The findings suggest that dietary leucine may delay early insulin signaling without affecting muscle's ability to uptake glucose.
