Liraglutide reduces clozapine-related energy problems in rat brains by protecting mitochondria

Abstract

Chronic treatment with clozapine (20 mg/kg) impaired cognitive function in rats and induced neuronal apoptosis.

  • Clozapine treatment was associated with cognitive deficits and increased neuronal cell death.
  • Electron microscopy revealed that clozapine damaged mitochondrial structure and function, leading to decreased energy production.
  • Clozapine treatment reduced levels of TCA cycle intermediates and increased oxidative stress in the hippocampus and prefrontal cortex.
  • Activation of the GLP-1 receptor by the agonist LIRA reversed some of the mitochondrial dysfunction caused by clozapine.
  • Inhibition of the GLP-1 receptor with Exendin9-39 negated the protective effects of LIRA.
