Long-lasting reduction of blood vessel leakiness in a hereditary swelling mouse model using targeted genetic editing

Abstract

AAV8-NG-ABE8e induced > 60% gene editing at the target site, resulting in over 85% reductions in serum kallikrein levels.

  • The intervention promoted exon 2 skipping of Klkb1 mRNA, leading to long-term effects sustained for at least 1 year.
  • Bradykinin-driven vascular hyperpermeability was reversed following treatment with AAV8-NG-ABE8e.
  • Inflammatory gene signatures were attenuated in response to the gene editing.
  • Prolongation of activated partial thromboplastin time (aPTT) occurred only when kallikrein levels fell below 2 μg/mL.
  • Lipid nanoparticles also achieved > 90% reductions in Klkb1 expression and serum kallikrein levels without significant long-term toxicity.
