Reduced Adaptability in the D2-Controlled Reward Pathway May Increase Cocaine Craving

Abstract

Increasing activity in D1-MSNs or decreasing activity in D2-MSNs augmented cue-induced reinstatement of cocaine-seeking behavior in a mouse model.

  • D1-MSNs promote motivated behaviors, while D2-MSNs inhibit them.
  • Chemogenetic regulation showed that both D1- and D2-MSNs innervate the ventral pallidum with GABAergic projections.
  • Inhibition of ventral pallidal neurons blocked the increased reinstatement caused by changes in D1- or D2-MSN activity.
  • GABAergic long-term depression (LTD) was lost in D2-MSN synapses after cocaine self-administration, but not in D1-MSNs.
  • Increased enkephalin activity on μ opioid receptors occluded LTD in D2-MSNs, indicating a potential mechanism for the observed changes.
