Ventral Pallidum as the Main Target of Accumbens D1 Neurons Driving Cocaine Seeking

Abstract

Approximately 70% of D1 dopamine receptor-expressing neurons in the nucleus accumbens project to both the ventral pallidum and ventral mesencephalon.

  • D1 dopamine receptor-expressing neurons consist of two populations that are segregated by differentially expressed proteins.
  • A large majority of D1-MSNs in the nucleus accumbens collateralize to both the ventral pallidum and the ventral mesencephalon.
  • More than 90% of D1-MSNs projecting to the ventral mesencephalon also collateralize to the ventral pallidum.
  • In contrast, less than 10% of dorsal striatal D1-MSNs collateralize to both the globus pallidus and ventral mesencephalon.
  • Inhibition of D1-MSNs in the ventral pallidum, but not in the ventral mesencephalon, blocks cue-induced cocaine seeking.
  • These findings suggest that only D1-MSN projections to the ventral pallidum are necessary for responding to cocaine-conditioned cues.
