Abstract

In a study of 10 normal, healthy males, 8 of 10 individuals stabilized their sleep-wake cycle to 24 hours with the first administration of melatonin.

  • Melatonin administration may phase shift sleep-wake cycles in humans.
  • In the first treatment phase, 9 of 10 subjects exhibited free running with placebo, while melatonin stabilized the sleep-wake cycle in 8 of 10.
  • Melatonin resulted in phase advances in sleep-wake cycles for some individuals and irregular sleep patterns for 2 subjects.
  • While melatonin influenced sleep patterns, core temperature rhythms continued to free run in 4 subjects.
  • Maximum phase advances in core temperature occurred when melatonin was administered approximately 2 hours after the peak temperature.
