Abstract

A metabolomic clock was highly predictive of chronological age (r=0.92) based on analysis of 3,686 plasma samples from 2,295 participants.

  • Certain metabolites, including N2,N2-dimethylguanosine and zeta-carotene, are associated with chronological age, frailty, and mortality.
  • The noradrenergic metabolite 3-methoxy-4-hydroxyphenylglycol sulphate and the oligosaccharide sialyllactose show strong associations with both age and mortality.
  • Metabolomic age acceleration is correlated between study visits (r > 0.6).
  • Each standard deviation increase in metabolomic age acceleration (approximately 5 years) is linked to a 43% higher mortality risk and a 27% higher risk of mild cognitive impairment.
