Abstract

3-Oxo-lithocholic acid (3-oxo-LCA) significantly reduced tumor burden in a genetically engineered mouse model of colorectal cancer.

  • 3-Oxo-LCA activated FXR signaling, which is linked to reduced growth of colorectal cancer cell lines.
  • In vitro, 3-oxo-LCA suppressed proliferation of intestinal stem cells in both mouse and patient-derived organoids.
  • In a genetically engineered mouse model, 3-oxo-LCA enhanced gut barrier function and decreased tumor initiation.
  • 3-Oxo-LCA inhibited tumor progression and promoted apoptosis in syngeneic and xenograft mouse models.
  • These findings suggest that 3-oxo-LCA may impact colorectal cancer tumorigenesis and progression through modulation of epithelial cell behavior.
