ThermoCas9 adds methylation sensitivity to genome editing by cutting unmethylated PAM targets while being inhibited by methylated cytosines.

Evidence

Biochemical assays, 2.8 A and 2.2 A cryo-EM structures, and human cell-line editing experiments characterized ThermoCas9 binding, cleavage, and methylation-sensitive targeting.

Caveat

The disease-targeting claim is based on cell-line editing and hypomethylated breast-cancer signature genes, not patient treatment or clinical outcomes.
