Nine bile acid-derived sterols were engineered to assess their impact on mRNA-lipid nanoparticles, revealing that sterol structure governs formulation behavior and organ expression profiles.

  • Formulation behaviors of mRNA-lipid nanoparticles correlate with structural features of bile acid-derived sterols as determined by all-atom molecular dynamics simulations.
  • Substituting cholesterol with these sterols reduced hepatic expression and favored spleen-targeted organ expression, which is important for immune response.
  • CA-20 lipid nanoparticles enhanced antigen-specific immune responses, including improved memory features, while demonstrating a favorable safety profile.
