An AI-designed lipid nanoparticle improved cartilage-selective mRNA delivery and supported chondrocyte gene editing with cartilage protection in mouse osteoarthritis models.

Evidence

This preclinical platform study used a multiobjective AI model to design ionizable lipids, then showed that K9 LNPs achieved over 90% transfection in mouse joint chondrocytes and 13.5-fold higher knee-to-liver selectivity than SM-102, with chondrocyte-specific Mmp13 editing and sustained cartilage protection in osteoarthritis mice.

Caveat

Because the results come from AI-guided nanoparticle optimization and mouse cartilage models, tissue selectivity, safety, and efficacy in humans remain unproven.
