Measuring nine GLP-1 receptor agonist drugs together using a high-precision chemical analysis method and applying it to medicine

Abstract

A new multiplexed liquid chromatography-high-resolution mass spectrometry method effectively identifies and quantifies nine peptide-based glucagon-like peptide-1 receptor agonists.

  • The method provides excellent resolution with baseline separation for most peptides within a 20-minute runtime.
  • Mass accuracy ranged from 0.0 to 0.5 ppm, with fragment ion coverage between 76% and 100%.
  • Linearity was demonstrated with a correlation coefficient (r²) of 0.995 or higher across a concentration range of 4-235 ng/mL.
  • Limit of detection (LOD) values were between 0.44 and 2.06 ng/mL, while limit of quantification (LOQ) values ranged from 8.29 to 11.74 ng/mL.
  • The method yielded mean recoveries of 113.7% to 118.4% when applied to marketed formulations.
