Modified GLP1R activators favor G protein signaling by shifting a specific outer receptor loop

Abstract

The cryo-EM structure of the GLP1-Y11-GLP1R-Gs complex was resolved at 2.64 Å.

  • GLP1R is a key target for treating type 2 diabetes and obesity.
  • G protein-biased GLP1R agonists show preferential signaling compared to the reference ligand.
  • Outward displacement of extracellular loop 3 (ECL3) is linked to G protein signaling bias.
  • Ac-GLP1 and Ac-semaglutide alter receptor trafficking and reduce beta-arrestin recruitment.
  • Ac-semaglutide effectively lowers glucose levels in diet-induced obese mice up to three days after administration.
