Brain and Mental Health Effects of GLP-1 Receptor Agonists in Adults with Overweight or Obesity

Updated

Abstract

Fourteen studies were included in the review, focusing on neuropsychiatric outcomes associated with GLP-1 receptor agonists in adults with overweight or obesity.

  • Randomized studies showed no consistent worsening of depressive symptoms or imbalance in suicide-related outcomes.
  • Observational studies reported a wide range of hazard ratios (HRs) for depression, from 0.63 to 2.95, indicating variability across different populations.
  • For suicide-related outcomes, observational HRs ranged from 0.27 to 2.06, while randomized studies indicated that suicidal ideation or behavior was uncommon.
  • Anxiety-related findings were inconsistent, and manic or hypomanic symptoms were not systematically evaluated.
  • The certainty of evidence was assessed as low for depressive symptoms and very low for anxiety and suicide-related outcomes.
