An improved system for precise gene silencing using CRISPR technology

The optimized transcriptional repressor dCas9-ZIM3-NID-MXD1-NLS achieves gene knockdown performance exceeding levels observed with traditional CRISPRi by an average of 40%.

  • Truncating the established repressor domain MeCP2 enhances CRISPRi gene knockdown performance significantly.
  • The ultra-compact NCoR/SMRT interaction domain (NID) is particularly effective, improving performance compared to canonical MeCP2 subdomains.
  • Combinatorial libraries of multi-domain repressors led to the discovery of four new repressor fusions.
  • Attaching a carboxy-terminal nuclear localization signal (NLS) improves gene knockdown efficiency by an average of ∼50%.
  • Validation across multiple cell lines and sgRNA targets confirms the superior capabilities of the developed dCas9-ZIM3-NID-MXD1-NLS system.
