Expression of NLRP3 in peritoneal macrophages varies with time of day and is regulated by NR1D1.

  • The activity of the NLRP3 inflammasome complex exhibits a circadian rhythm influenced by NR1D1.
  • Macrophages lacking NR1D1 showed altered expression of NLRP3 and reduced production of inflammatory cytokines IL1B and IL18.
  • Mice with disrupted Nr1d1 experienced more severe inflammation and liver failure compared to control mice.
  • Activation of NR1D1 using SR9009 decreased NLRP3 expression and cytokine secretion in macrophages.
  • Mice treated with SR9009 had less severe liver failure and longer survival times than those given a saline control.
