Watching structural changes during enzyme activation in different types of IscB and CRISPR-Cas9

Updated

Abstract

Stepwise nuclease activation mechanisms of diverse IscB and Cas9 nucleases have been visualized using cryo-electron microscopy and high-speed atomic force microscopy.

  • IscB family nucleases utilize two distinct strategies for HNH autoinhibition, with one strategy being the direct evolutionary precursor to Cas9 activation.
  • A conserved protein-DNA interaction stabilizes an intermediate activation state in both IscB and Cas9 enzymes.
  • Cas9 has developed an additional gating element to prevent premature activation before catalysis.
  • Structural dynamics of nuclease activation reveal both conserved and diversified mechanisms throughout the IscB-to-Cas9 lineage.
