Improved gGBE tool allows reliable G-to-Y DNA changes in mammal cells and zebrafish embryos

Updated

Abstract

A high-performance glycosidase guanine base editor (gGBE-PIGS) improved editing efficiency by up to 3.5-fold in mammalian cells and 13-fold in zebrafish embryos.

  • Base editing is a precise genome-editing tool that can introduce single-base substitutions.
  • Glycosidase-derived transversion editing has been challenging in mammalian cells.
  • The engineered gGBE-PIGS variant maintains high editing precision while enhancing efficiency.
  • A GFP reporter was developed to monitor and enrich for gGBE-edited cells and embryos.
  • Zebrafish models were successfully generated with mutations linked to human amyotrophic lateral sclerosis (ALS).
