Effects of the oral drug aleniglipron on people with overweight or obesity in a controlled clinical trial

Essence

Oral aleniglipron produced clinically relevant weight loss in adults with overweight or obesity over 36 weeks.

Evidence

A randomized, double-blind, placebo-controlled phase 2b trial assigned 230 adults to once-daily aleniglipron or placebo and found placebo-adjusted body-weight reductions of -8.2%, -9.8%, and -11.3% at 45, 90, and 120 mg.

Caveat

The trial was phase 2b with 36-week double-blind follow-up, an ongoing open-label extension, and treatment-related discontinuations of 10.4% across aleniglipron arms.
