Abstract
Peptide mapping achieved full sequence coverage of semaglutide and liraglutide using ultra-performance liquid chromatography-high-resolution mass spectrometry.
- Synthetic peptides for semaglutide and liraglutide may be comparable to original drugs produced via recombinant DNA technology.
- Masses of peptide fragments were identified with a mass error below 10 parts per million.
- The method allowed for the determination of the fatty acid chain modification site through in-source fragmentation.
- Combining peptide maps generated by Glu-C and chymotrypsin enabled comprehensive coverage of the peptide sequences.
- The developed mapping method may support the evaluation of sameness between generic synthetic peptides and approved peptide drug products.
