How Manufacturing and Mixing Affect the Quality and Properties of Follow-On GLP-1 Peptide Drugs

Abstract

A total of 16 injectable and 8 oral follow-on products of semaglutide were analyzed for purity and stability.

  • Follow-on injectable semaglutide products displayed new impurities, including high molecular weight proteins, trace metals, and residual solvents.
  • Several commercialized oral semaglutide products contained significantly lower amounts of the drug than advertised on their labels.
  • Dissolution tests revealed varying release profiles for semaglutide and its absorption enhancer, potentially affecting bioavailability.
  • Neoepitopes suggesting potential immunogenicity were identified in both drug substances and products of follow-on semaglutide.
  • Liraglutide follow-on products showed increased fibrillation and reduced physical stability compared to the originator formulations.
