Abstract

Thirteen known impurities were identified from the degradation of semaglutide (SGL) under various stress conditions.

  • The degradation process of SGL was influenced by factors such as pH, temperature, buffer species, and molarity.
  • Thermal stress studies were conducted at temperatures of 25 °C, 40 °C, 60 °C, and 80 °C over varying durations.
  • Six impurities consistently formed across all testing conditions, indicating their relative stability compared to others.
  • Some impurities were absent across all pH conditions but emerged when water was used as a solvent.
  • The study suggests that pH is a crucial factor affecting the thermal stability of SGL.
  • The findings could assist in optimizing formulation processes and improving the stability of SGL during storage and transportation.
