Measurement of leftover plasmid DNA and viral promoter sequences in Pfizer/BioNTech and Moderna mRNA COVID-19 vaccines from Ontario, Canada

Updated

Abstract

Essence

This vial analysis found measurable residual DNA in Pfizer-BioNTech and Moderna COVID-19 modRNA vaccines, with SV40 promoter-enhancer sequences detected only in Pfizer samples tested here.

Evidence

This is a laboratory measurement study of 32 vials from 16 lots using Qubit fluorometry, qPCR, RNase A treatment, and one Oxford Nanopore sequencing run to quantify total and target-specific DNA.

Caveat

The study examined a limited vial sample and method-dependent results diverged, with all Moderna vials within regulatory limits by qPCR but not by fluorometry.
