Poricoic acid A reduces colitis symptoms in mice by controlling cell recycling and preventing gut cell death

Updated

Abstract

PAA treatment at 10 mg/kg significantly alleviated weight loss and colon shortening in mice with DSS-induced colitis.

  • PAA reduced disease activity index scores and levels of colonic IL-1β and TNF-α.
  • Histological analysis showed decreased colonic crypt damage and inflammatory cell infiltration with PAA treatment.
  • PAA increased goblet cell counts in the colon compared to the DSS group.
  • In Caco-2 cells, PAA inhibited the downregulation of tight junction proteins caused by DSS.
  • PAA treatment increased Bcl-2 expression and the LC3-II/I ratio while decreasing Bax and cleaved caspase-3 levels.
  • The mechanism suggests PAA may target the AMPK/mTOR pathway to activate autophagy and inhibit apoptosis.
