The optimized prime editing system achieved an average desired editing frequency of 80.83% in cucumber.

  • Significant improvements in transformation efficiency were observed by introducing spectinomycin in cucurbits.
  • Enhanced PE protein expression was achieved using the tomato elongation factor 1-alpha promoter.
  • The incorporation of the Csy4 ribonuclease facilitated the processing of pegRNAs.
  • Csy4-PE6d achieved editing frequencies of up to 100% at certain targeted loci.
  • Homozygous edits were generated in 36.43% of transgenic cucumber lines.
