Psychedelics long-lastingly increase reward response in male rats

Abstract

Reward responsiveness significantly increased after administration of psilocybin (0.1-1 mg/kg) or ketamine (10 mg/kg) in rats (n=84).

  • Psilocybin and ketamine produced both acute and sustained increases in reward responsiveness lasting 24 hours post-administration.
  • The increase in reward responsiveness due to psilocybin was blocked in a dose-dependent manner by the 5-HT2A receptor antagonist volinanserin.
  • DMT and (±)-DOI also produced significant acute increases in reward responsiveness, but these effects did not persist 24 hours later.
  • Neither the non-psychedelic 5-HT2A agonist lisuride nor the SSRI fluoxetine showed positive effects on reward responsiveness.
  • The findings suggest that the enhancement of reward responsiveness by psychedelics may vary in duration and depends, at least in part, on the 5-HT2A receptor.
