Abstract
Renal OTR mRNA levels decreased by more than 3-fold at term in rats.
- Two major OTR messenger RNA bands were detected in renal extracts, measuring 6.7 and 4.8 kilobases.
- In contrast to the uterus and pituitary, which showed increased OTR mRNA levels at term, renal levels fell significantly.
- Estrogen treatment resulted in increased renal OTR mRNA levels but produced shorter transcripts (3.6 and 3.2 kb) compared to control kidneys.
- The E2-induced transcripts shared the same coding region as the longer transcripts but had a shortened 3'-untranslated region.
- Increased renal OT-binding sites were observed following estrogen treatment, indicating the potential functionality of the shorter OTR transcripts.
- The regulation of OTR gene expression appears to differ between the uterus and kidneys, suggesting a role for oxytocin in both reproduction and sodium balance.
