REV-ERBα slows bone-breakdown cell formation and helps prevent bone loss around teeth

Abstract

A novel Periodontitis Circadian Rhythm Score (PeriCRS) model was established using machine learning on transcriptomic data.

  • Circadian rhythm disruption may influence periodontitis by affecting molecular clock genes involved in bone homeostasis.
  • Key molecular clock genes associated with alveolar bone destruction were identified through bioinformatics analysis.
  • The expression of the molecular clock gene Rev-erbα was found to be abnormal in inflammatory periodontal tissue in mice.
  • Activation of REV-ERBα with the agonist SR9009 could reduce osteoclast differentiation and related factors.
  • In vivo experiments indicated that SR9009 may mitigate alveolar bone loss caused by periodontitis.
