Single-cell CRISPR tests show enhancers mostly increase gene activity by multiplying effects

Abstract

A total of 46,166 enhancer pairs were tested for interactions with genes using the GLiMMIRS framework.

  • Enhancer effects are observed to combine multiplicatively.
  • Only 31 enhancer pairs showed significant interactions, with none from the high-confidence set.
  • 20 of the significant interactions were influenced by outlier expression values.
  • Further analyses suggest that enhancer interactions may be uncommon or have small effects that are hard to detect.
