Creating circular RNAs that produce long-lasting diabetes drugs without scars

Abstract

Scarless circRNAs encoding GLP-1 or its derivative demonstrated potent and sustained hypoglycemic effects in diabetic mice comparable to Semaglutide over a 7-day period.

  • The SLPIE system enabled efficient circularization of RNA with minimized immunogenicity.
  • Systematic optimization revealed the EV-B107_IRES to be highly compatible with the SLPIE strategy, enhancing translational efficiency.
  • Incorporation of 5-7% m⁶A modification during RNA synthesis improved translation when using lipid nanoparticles.
  • The synthesized circRNAs showed significant anti-obesity effects alongside their hypoglycemic actions in diabetic mice.
  • A scalable production platform was established using an RNase III-deficient E. coli strain.
